      目的 研究红树药用植物红榄李内生真菌Penicillium sp. HLLG-122的次级代谢产物及生物活性。方法 采用大米固体发酵培养,综合运用正相硅胶柱色谱、反相C18柱色谱、半制备高效液相色谱法等现代色谱分离技术纯化单体化合物,运用核磁共振波谱(NMR)、质谱(MS)等方法并结合文献报道数据对化合物进行结构鉴定。结果 从内生真菌Penicillium sp. HLLG-122中共分离鉴定得到11个杂萜类化合物,分别鉴定为:eupeniacetal B(1)、talaromynoid A(2)、talaromynoid C(3)、talaromynoid F(4)、talaromynoid H(5)、miniolutelide C(6)、purpurogenolide C(7)、preaustinoid A7(8)、berkeleyone C(9)、preaustinoid C(10)和berkeleyone A(11)。细胞毒活性测试结果表明化合物10对HepG2和MCF-7癌细胞系具有中等的细胞毒活性,半抑制浓度(IC50)分别为15.1和35.5 μmol/L;化合物11对MCF-7癌细胞系显示有中等的细胞毒活性,IC50为25.2 μmol/L。结论 化合物1~11均从红榄李来源真菌中分离获得,其中化合物2~5为首次从青霉菌属中分离得到。化合物10和11对HepG2和MCF-7癌细胞系具有中等的细胞毒活性。
English Summary:
      Objective The secondary metabolites and biological activities of the endophytic fungus Penicillium sp. HLLG-122 from the mangrove Lumnitzera Littorea were investigated. Methods The endophytic fungi Penicillium sp. HLLG-122 was cultivated in rice medium. The monomer compounds were purified by normal and reverse phase silica gel and semi-preparative HPLC column chromatography, and the structures of the compounds were identified by HR-ESI-MS, NMR spectroscopic data and comparison with literature data. Results Eleven meroterpenoid derivatives, eupeniacetal B (1), talaromynoid A (2), talaromynoid C (3), talaromynoid F (4), talaromynoid H (5), miniolutelide C (6), purpurogenolide C (7), preaustinoid A7 (8), berkeleyone C (9), preaustinoid C (10) and berkeleyone A (11) were isolated and identified from the endophytic fungus Penicillium sp. HLLG-122. The results of cytotoxic activity test showed that compound 10 exhibited moderate cytotoxicity against HepG2 and MCF-7 cell lines with the IC50 values of 15.1 and 35.5 μmol/L, respectively; Compound 11 exhibited moderate cytotoxicity against MCF-7 cell line with the IC50 value of 25.2 μmol/L. Conclusion Compounds 1?11 were isolated from the endophytic fungus of Lumnitzera Littorea, and the compounds 2?5 were isolated from Penicillium for the first time. Compounds 10 and 11 showed moderate cytotoxicity against HepG2 and MCF-7 cell lines.
