      目的 利用模式生物斑马鱼,对筛选具有抑制Wnt/β-catenin信号通路活性的海洋来源天然化合物进行研究。方法 在受精后6小时(6 hours post fertilization,6 hpf)加入BIO及不同浓度的海洋来源化合物,通过观察48 hpf斑马鱼胚胎眼睛发育情况,检测海洋来源天然化合物对斑马鱼眼部表型的营救作用,同时,本研究选择效果较好的化合物1#、6#、9#、11#在细胞水平进行进一步探究。结果 部分海洋来源天然化合物能够不同程度地营救BIO处理导致的斑马鱼胚胎眼部发育缺陷的表型,同时在细胞水平,化合物与BIO共处理能抑制BIO诱导的Wnt信号通路的异常激活。
English Summary:
      Abstract: Objective To screen Marine natural compounds with inhibitory activity of Wnt/β-catenin signaling pathway by using model biological zebrafish. Methods Biofertilization and different concentrations of marine-derived compounds were added to 6 hpf (6 hours post fertilization, 6 hpf). The rescue effects of marine-derived natural compounds on ocular phenotypes of zebrafish were examined by observing the eye development of 48 hpf embryos. In this study, compounds 1#, 6#, 9# and 11# with good effects were selected for further exploration at the cellular level. Results Some natural compounds of Marine origin could rescue the phenotype of eye defects caused by BIO treatment to varying degrees. At the cellular level, co-treatment of the compounds with BIO could inhibit Bio-induced abnormal activation of Wnt signaling pathway.
