目的 通过淘选获得高亲和力和特异性的抗CD38纳米抗体,为以海洋来源毒素为基础的抗体偶联药物提供新的候选抗体分子。方法 表达并纯化人CD38蛋白胞外段,进行羊驼免疫,进而构建羊驼纳米抗体噬菌体展示文库。从噬菌体展示文库中筛选抗CD38特异性纳米抗体,利用ELISA和流式细胞术等技术对纳米抗体进行表征。结果 构建的免疫文库库容为2.17 × 109 cfu/mL,插入率100%,共获得22株CDR3不同的抗CD38纳米抗体,其中4株纳米抗体能高亲和力结合CD38,3株纳米抗体能与Daudi细胞结合。结论 成功获得了亲和力高、特异性的CD38纳米抗体,为海洋毒素类抗体偶联药物提供了新的候选抗体分子,有望替代传统抗体用于多发性骨髓瘤的治疗。

Abstract: Objective The high affinity and specific anti-CD38 nanobodies were obtained by panning, which provided new nanobody candidates for ADC drugs based on marine toxins. Methods The extracellular segment of human CD38 protein was expressed and purified to immunize alpaca, then the phage library was constructed. Anti-CD38 specific nanobodies were screened from phage display library and characterized by ELISA and flow cytometry. .Results The immune library was constructed with a capacity of 2.17×10^9 cfu·mL-1 and an insertion rate of 100%. 22 anti-CD38 nanobodies with different CDR3 were selected, among which 4 nanobodies could bind CD38 with high affinity and 3 nanobodies could bind to Daudi cells. Conclusion CD38 nanobodies with high affinity and strong specificity were successfully obtained, which provided new nanobody candidates for ADC drugs based on marine toxins, which are expected to replace traditional antibodies for the treatment of Multiple Myeloma.
