      摘要:目的 对来源于西北太平洋水深5000米以下沉积物分离的真菌Aspergillus clavatonanicus次级代谢产物及其抗氧化活性进行研究。方法 利用薄层色谱、正向硅胶柱层析、高效液相色谱等分离手段对真菌Aspergillus clavatonanicus次级代谢产物进行分离、纯化;运用核磁共振波谱、质谱等方法,通过与报道数据进行对比,对化合物的化学结构进行鉴定,并用DPPH自由基清除实验,对化合物抗氧化活性进行测试。结果 从真菌Aspergillus clavatonanicus发酵产物中分离得到8个吲哚喹唑啉类生物碱,分别为tryptoquivaline (1)、nortryptoquivaline (2)、aspertoryadin C (3)、fiscalin B (4)、deoxytryptoquivaline (5)、aspertoryadin C (6)、fiscalin C (7)、epi-fiscalin C (8),其中化合物3表现出一定的抗氧化活性,其半抑制浓度(IC50)为7.6 μg·mL?1。结论 从真菌Aspergillus clavatonanicus发酵产物中分离得到8个吲哚喹唑啉类生物碱,其中化合物3具有一定的抗氧化活性,可为抗氧化剂的研究提供一定的数据支持。
English Summary:
      Abstract: ObJective Secondary metabolites and their antioxidant activities were investigated in the fungus Aspergillus clavatonanicus isolated from sediments in the Northwest Pacific Ocean at water depths up to 5000 meters. Methods Separation and purification of secondary metabolites of the fungus Aspergillus clavatonanicus using thin-layer chromatography,forward silica gel column chromatography and HPLC. Characterization of the chemical structures of the compounds using NMR,MS and by comparing them with the reported data. They were also test their antioxidant activity using the DPPH free radical scavenging assay. Results Eight indole quinazoline alkaloid compounds,were isolated from the fermentation products of the fungus Aspergillus clavatonanicus. They were identified as tryptoquivaline (1)、nortryptoquivaline (2)、aspertoryadin C (3)、fiscalin B (4)、deoxytryptoquivaline (5)、aspertoryadin C (6)、fiscalin C (7)、epi-fiscalin C (8). Compound 3 showed antioxidant inhibitory activity with IC50 value of 7.6 μg·mL?1. Conclusion Eight indole quinazoline alkaloid compounds were isolated from the secondary metabolites of the fungus Aspergillus clavatonanicus. Compound 3 has some antioxidant activity and can provide some data support for antioxidant research.
