      目的:对从蠕食性橡果螺(Conus quercinus)获得的全新的芋螺毒素序列Qc-VB-01进行合成和抗菌活性评价。方法:利用固相肽合成方法合成线性肽,2,2-二硫二吡啶氧化法连接二硫键,以微量肉汤稀释法测试该多肽对5种致病好氧菌的最低抑菌浓度(MIC)。结果:成功合成芋螺毒素Qc-VB-01及其突变体M10A、M16A,利用质谱验证其分子量和分析液相证明其纯度大于95%。发现Qc-VB-01具有一定抗菌活性,尤其对于金黄色葡萄球菌CMCC(B) 26003效果显著。甲硫氨酸替换为丙氨酸后,不影响活性结果。结论:Qc-VB-01是具有一定抗菌活性的芋螺毒素,结构新奇简单而抗菌效果明显。
English Summary:
      Objective: To synthesize a novel conotoxin Qc-VB-01 obtained from Conus quercinus and evaluate its antibacterial activity. Methods: The linear peptide was synthesized by solid phase peptide synthesis method, the disulfide bond was linked by 2, 2-disulfide dipyridine oxidation method, and the minimum inhibitory concentration (MIC) of the peptide against 5 pathogenic aerobic bacteria was tested by microbroth dilution method. Results: Conotoxin Qc-VB-01 and its mutants M5A and M11A were synthesized successfully. Their molecular weight was verified by mass spectrometry and their purity was more than 95% by liquid phase analysis. It was found that Qc-VB-01 had good antibacterial activity, especially against Staphylococcus aureus CMCC(B) 26003. The replacement of methionine with alanine did not affect the activities of these mutants. Conclusion: Qc-VB-01 is a antibacterial peptide with a novel and simple structure.
