| 目的 对一株深海来源真菌Coniochaeta sp. PS09-A2-1的次级代谢产物进行研究。方法 运用硅胶柱层析、反相C18硅胶(ODS)柱层析、Sephadex-LH20凝胶柱层析、半制备高效液相色谱(HPLC)等方法对Coniochaeta sp. PS09-A2-1的大米发酵提取物进行分离纯化;通过核磁共振谱(NMR)及质谱(MS)等波谱学方法进行结构解析,通过X 射线单晶衍射实验确定新化合物的绝对构型,并测定化合物的抗炎及抗氧化活性。结果 从Coniochaeta sp. PS09-A2-1大米发酵提取物中分离鉴定了2个蒽醌类化合物(2和5)、2个 α-吡喃酮类化合物(3和6)、1个蒽醌-α-吡喃酮二聚体化合物(4)和1个萜类化合物(1),其中包括4个新化合物(1-4)和2个已知化合物(5和6)。抗炎活性评价结果表明,新化合物4在脂多糖(LPS)刺激的BV-2细胞的模型中表现出抗炎活性;抗氧化活性评价结果表明,化合物4具有显著的 2,2′- 联氮基双-(3-乙基苯并噻唑啉-6-磺酸)(ABTS)自由基清除活性。结论 化合物1~6均从深海来源真菌分离获得,其中化合物1~4为首次从锥毛壳菌属中分离得到。化合物4对在脂多糖(LPS)刺激的BV-2细胞的模型中表现出抗炎活性并且具有显著的2,2′-联氮基双-(3-乙基苯并噻唑啉-6-磺酸)(ABTS)自由基清除活性。 |

| Objective To study the secondary metabolites of a fungus Coniochaeta sp. PS09-A2-1. Method The compounds were isolated by Silica gel, reverse-phase C18 Silica gel (ODS), Sephadex LH-20 gel column chromatography, and semi-preparative high-performance liquid chromatography (HPLC) from the organic extract of its rice culture. Their structures were identified by comprehensive NMR and MS spectroscopic methods as well as comparison with previously related literature, and the absolute configurations were established by single crystal X-ray diffraction. The anti-inflammatory and antioxidant activities of the compounds were evaluated. Result Two anthraquinones (2 and 5), two α-pyrones (3 and 6), one anthraquinone-α-pyrone dimer (4), and one terpenoid (1) were isolated and identified from the crude extract of Coniochaeta sp. PS09-A2-1, including four new compounds (1-4) and two known compounds (5 and 6). Compound 4 exhibited anti-inflammatory activity in the LPS-stimulated BV-2 cell model and antioxidant activity tests indicated that compound 4 possesed significant 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity.Conclusion Compounds 1–6 were all isolated from deep-sea-derived fungi, among which compounds 1–4 were obtained from the genus Coniochaeta sp. for the first time. Compound 4 exhibited anti-inflammatory activity in the LPS-stimulated BV-2 cell model and demonstrated significant 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity. |
