      目的 对中国南海涠洲岛来源刺柳珊瑚 Echinogorgia rebekka 中甾体类次级代谢产物进行分离纯化,结构鉴定和体外生物活性测试。方法 利用硅胶柱层析、凝胶柱层析、半制备高效液相色谱等手段进行分离纯化;综合运用质谱、一维和二维核磁共振波谱、旋光、X-射线单晶衍射等方法,并对比相关文献数据对单体化合物进行结构鉴定;建立LPS诱导的巨噬细胞炎症模型进行活性评价。结果 从E. rebekka刺柳珊瑚中分离鉴定8个甾体类化合物,包括1个新化合物Rebekksteroid A (1)和7个已知化合物Caerulsteroid A (2),Menellsteroid A (3),Cholestane-3β, 5α, 6β-tetrol (4),Cholest-5-ene-3β,7α-diol (5),Cholesta-5-en-3β,7β,19-triol (6),Cholestane-1β, 3β, 5α, 6β-tetraol (7),3β-hydroxycholesta-5-en-7-one (8)。结论 化合物1为新化合物,化合物2,5和8首次从该种属中分离得到。体外活性结果显示,化合物3和5具有中等抗炎活性。
English Summary:
      Objective To isolate and purify steroidal secondary metabolites from the gorgonian coral Echinogorgia rebekka collected from Weizhou Island in the South China Sea, elucidate their structures and evaluate their in vitro bioactivities. Methods The metabolites were separated and purified using silica gel column chromatography, gel filtration, and semi-preparative HPLC. Structural elucidation of the isolated compounds was performed by comprehensive analysis of MS, 1D and 2D NMR spectroscopy, optical rotation, and single-crystal X-ray diffraction data, combined with comparison to reported literature. In vitro anti-inflammatory activity was assessed using an established LPS-induced RAW264.7 cell assay. Results Eight steroidal compounds were isolated and identified from E. rebekka, including one new compound: Rebekksteroid A (1), and seven known compounds: Caerulsteroid A (2), Menellsteroid A (3), cholestane-3β,5α,6β-triol (4), cholest-5-ene-3β,7α-diol (5), cholesta-5-en-3β,7β,19-triol (6), cholestane-1β,3β,5α,6β-tetraol (7), and 3β-hydroxycholesta-5-en-7-one (8). Compounds 2, 5, and 8 were obtained from this genus for the first time. Conclusion Compound 1 is a new steroid, and compounds 2, 5, and 8 are isolated from the genus Echinogorgia for the first time. In vitro assays revealed that compounds 3 and 5 exhibited moderate anti-inflammatory activity.
