| 目的 建立一种高效提取海洋药物先导物CA的超声辅助乙醇提取工艺,为其规模化制备提供参考。方法 以海洋链霉菌诱变菌株891-B6的干燥菌丝体为研究对象,以CA得率为指标,单因素试验分别考察了超声功率、提取温度、液料比(mL:g)、提取时间和提取次数对其影响,进一步采用响应面Box-Behnken设计优化了CA的提取工艺条件。结果 CA最佳提取工艺条件为液料比360 mL·g?1、提取温度50 ℃、超声功率100 W、提取时间30 min、提取3次,在此提取条件下,CA得率最高,达108.3±3.6 mg·g?1,与回归模型预测值接近,且比传统加热提取法和索式提取法效率更高。结论 超声辅助乙醇提取CA工艺绿色、高效,具有重要的应用价值。 |

| Objective To establish an ultrasonic-assisted extraction (UAE) process for efficient preparation of marine drug candidate CA using ethanol as solvent, providing a key reference for its large-scale production. Methods Using dried mycelia of the marine-derived CA-producing mutant Streptomyces sp. 891-B6 as raw material, a single-factor experimental design was employed to explore the effects of UAE factors including ultrasonic power, extraction temperature, liquid-to-solid ratio (mL:g), extraction time and number of extraction cycles on CA yield. And these factors were further optimized using response surface methodology (RSM) with a Box-Behnken design (BBD) Results The optimal CA extraction conditions were determined as follows: liquid-to-solid ratio of 361 mL·g-1, extraction temperature of 50 °C, ultrasonic power of 100 W, extraction time of 30 min, and 3 extraction cycles. Under these conditions, the CA yield reached up to a maximum of 108.3 ± 3.6 mg·g-1, which was very close to the value predicted by the RSM regression model. Compared with traditional heating extraction and Soxhlet extraction methods, UAE is more efficient. Conclusion UAE is a simple, green and efficient method to extract CA from mycelia of strain 891-B6. |
