      目的 制备LY104雾化吸入液并研究其对卵清蛋白(Ovalbumin,OVA)诱导的小鼠哮喘的治疗作用。方法 制备LY104吸入液并检验其在高温灭菌、高温高湿和强光照射10 d内的稳定性。对Balb/c小鼠建立OVA诱导哮喘模型,分为空白组、模型组、阳性组(地塞米松1.0 mg/kg)、0.03 mg/kg LY104雾化吸入液组、0.1 mg/kg LY104雾化吸入液组。间隔给药3次,检测肺功能指标、肺泡灌洗液炎症因子和炎症细胞数量,并对肺组织进行苏木精伊红(HE)染色观察,考察LY104吸入液对小鼠哮喘的治疗效果。结果 LY104吸入液处方LY104、葡萄糖和磷酸二氢钠/磷酸氢二钠,pH约为7,渗透压约为744Pa;在高压蒸汽灭菌、高温和光照环境中具有良好的稳定性。LY104吸入液能够显著改善哮喘小鼠肺功能(P<0.000 1),明显降低OVA诱导的哮喘小鼠BALF中TNF-α、IL-6等炎症因子的水平(P<0.001或P<0.000 1),降低小鼠肺泡灌洗液(Bronchoalveolar Lavage Fluid,BALF)中炎症细胞数量(P<0.001或P<0.000 1),缓解小鼠肺部炎症细胞浸润、支气管壁增厚和肺泡壁结构的破坏。结论 LY104吸入液理化性质合格,稳定性良好,对OVA诱导的哮喘小鼠具有显著的治疗作用。
English Summary:
      Objective To prepare LY104 nebulized inhalation solution and investigate its therapeutic effect on ovalbumin (OVA)-induced asthma in mice. Methods The LY104 inhalation solution was prepared, and its stability was tested under conditions of high-temperature sterilization, high temperature and high humidity, and strong light irradiation for 10 days. Balb/c mice were used to establish an OVA - induced asthma model and then divided into the following groups: blank group, model group, positive (dexamethasone 1.0 mg/kg) group, 0.03 mg/kg LY104 nebulized inhalation solution group, and 0.1 mg/kg LY104 nebulized inhalation solution group. The mice were administered the drug three times at intervals. Pulmonary function indicators, inflammatory factors and the number of inflammatory cells in bronchoalveolar lavage fluid (BALF) were detected, and lung tissues were stained with hematoxylin and eosin (HE) for observation to evaluate the therapeutic effect of LY104 inhalation solution on asthma in mice. Results The formulation of LY104 inhalation solution consisted of LY104, glucose, and sodium dihydrogen phosphate/disodium hydrogen phosphate, with a pH value of approximately 7 and an osmotic pressure of approximately 744 Pa. It showed good stability under high-pressure steam sterilization, high-temperature, and light-exposure conditions. LY104 inhalation solution significantly improved the pulmonary function of asthmatic mice (P<0.000 1), notably reduced the levels of inflammatory factors such as TNF-α and IL-6 in the BALF of OVA-induced asthmatic mice (P<0.001 or P<0.000 1), decreased the number of inflammatory cells in the BALF of mice (P<0.001 or P<0.000 1), and alleviated inflammatory cell infiltration in the lungs, thickening of the bronchial wall, and destruction of the alveolar wall structure in mice. Conclusion The LY104 inhalation solution meets the requirements for physical and chemical properties, has good stability, and exerts a significant therapeutic effect on OVA - induced asthma in mice.
