目的 基于脂类药效成分,比较昆布类中药材海带与裙带菜的保护心血管作用。方法 收集10批次海带和12批裙带菜,采用改良的Bligh-Dyer法提取总脂,使用UPLC-Q Exactive-Orbitrap/MS非靶向分析极性脂质,采用GC-MS靶向分析总脂肪酸,利用化学模式识别法发现差异脂质,运用网络药理学预测心血管保护作用的可能途径及靶点。结果 海带总脂含量较低,仅为裙带菜的一半左右;两者脂类组成相似,主要差异在于优势分子种不同;两者均含有丰富的甘油糖脂和磷脂,但脂肪酰基构成在昆布中主要为饱和脂肪酸,在裙带菜中主要为多不饱和脂肪酸,与总脂肪酸分析结果一致。网络药理学提示,PLD信号通路和PI3K-AKT信号通路是海带和裙带菜保护心血管的共同通路;此外裙带菜还通过AGE-RAGE信号通路发挥作用。结论 昆布和裙带菜的脂类成分存在显著差异,这提示其可能影响了两者对心血管的保护作用,研究结果为临床用药提供一定的理论指导依据。 |

Objective Based on the pharmacodynamic material basis of lipids, the protective cardiovascular effects of Laminaria japonica and Undaria pinnatifida were compared. Methods Total lipids of ten batches of Laminaria japonica and twelve batches of Undaria pinnatifida were extracted by modified Bligh-Dyer method, UPLC-Q Exactive-Orbitrap/MS was used for global analysis of polar lipids and GC-MS was used for targeted analysis of total fatty acids. Chemical pattern recognition analysis was employed to find differential lipids. Network pharmacology was utilized to explore the possible pathways and target of cardiovascular protective effects. Results The total fat content of Laminaria japonica was lower, only about half of that of Undaria pinnatifida. The lipid composition of both was similar, while the dominant molecular species were different. Both of them are rich in glycerides and phospholipids. While the saturated fatty acyl chains are dominant in Laminaria japonica and the polyunsaturated fatty acyl chains are rich in Undaria pinnatifida, which is consistent with the results of total fatty acid analysis. Network pharmacology suggested that PLD signaling pathway and PI3K-AKT signaling pathway were common pathways for cardiovascular protection of Laminaria japonica and Undaria pinnatifida. In addition, Undaria pinnatifida also exerted effects through AGE-RAGE signaling pathway. Conclusion The lipid of Laminaria japonica and Undaria pinnatifida demonstrates significant differences, suggesting that these distinctions potentially influence their cardiovascular protective effect. The results of this study provide theoretical guidance for clinical medication. |
