目的 对1株来源北极Ny-?lesund地区的真菌Nectria sp. B-13的次级代谢产物进行研究。方法 采用减压柱色谱、正相硅胶柱色谱和高效液相色谱等多种色谱技术进行对该菌株的次级代谢产物进行分离纯化,运用核磁共振谱(NMR)、质谱(MS)和紫外光谱(UV)等波谱学方法并结合文献确定了化合物的平面结构。结果 从真菌Nectria sp. B-13的液体发酵物中分离鉴定出4个化合物:2-(4-hydroxy-4-methylpentyl)-2-methylbenzo[d][1,3]dioxole-5-carboxylic acid(1)、2-(5-hydroxy-4-methylpentyl)-2-methylbenzo[d][1,3]dioxole-5-carboxylic acid(2)、3-(2,5-dimethylbenzo[d][1,3]dioxol-2-yl)propanoic acid(3)、(7S)-(?)-10-hydroxysydonic acid(4),其中化合物1为新化合物,化合物2,3,4均是首次从该种属菌分离出来。对4个化合物进行生物活性测试,抗菌结果显示4个化合物的最大抑菌浓度(MIC)均大于64 μg/mL,无显著抗菌活性。 |

Objective Secondary metabolites from fungus Nectria sp. B-13 derived from the Ny-?lesund region of the Arctic were studied. Methods The secondary metabolites were isolated and purified using a variety of chromatographic techniques, including vacuum liquid chromatography, silica gel column chromatography, and high performance liquid chromatography (HPLC). The structures of the isolated compounds were determined by using spectroscopic methods such as nuclear magnetic resonance (NMR), spectroscopy (NMR), mass spectrometry (MS), and ultraviolet spectroscopy (UV), as well as by comparison the above data with the literatures. Results Four compounds were isolated and identified from the liquid fermentation of the fungus Nectria sp. B-13: 2-(4-hydroxy-4-methylpentyl)-2-methylbenzo[d][1,3]dioxole-5-carboxylic acid (1), 2-(5-hydroxy-4-methylentyl)-2-methylbenzo[d][1,3]dioxole-5-carboxylic acid (2), 3-(2,5-dimethylbenzo[d][1,3]dioxol-2-yl)propanoic acid (3), and (7S)-(?)-10-hydroxysydonic acid (4). Among them, compound 1 is new compound and compounds 2, 3, and 4 are all isolated for the first time from this genus of Nectria. These four compounds were tested for the antibacterial activity. Unfortunately, none of the four compounds showed antibacterial activity with the maximum inhibitory concentration (MIC) values > 64 μg/mL. |
