目的 研究海参内脏提取物对小鼠溃疡性结肠炎的治疗作用。方法 本研究通过探讨不同浓度的海参内脏提取物对葡聚糖硫酸钠(dextran sulfate sodium, DSS)诱导的溃疡性结肠炎小鼠疾病活动指数(disease activity index,DAI)、结肠组织病理、血清炎症因子和肠道微生物影响,评价其对溃疡性结肠炎的治疗作用。结果 海参内脏乙醇提取物(ethanol extract,EEx)和海参内脏水提取物(water extract,WEx)均可以剂量依赖性地缓解DSS引起的小鼠体质量下降以及DAI评分升高,缓解小鼠结肠的缩短,加快结肠组织损伤的恢复,降低粪便血红蛋白的含量,降低血清中促炎因子一氧化氮(NO)、肿瘤坏死因子-α(TNF-α)、白细胞介素1β(IL-1β)和IL-6的水平,增加抗炎因子IL-10的水平。同时,EEx和WEx可以降低肠道微生物中Blautia、Helicobacter和Erysipelatoclostridium等有害菌属的丰度,增加norank_f__Muribaculaceae、Lachnospiraceae_NK4A136_group等有益菌属的丰度,缓解DSS造成的有害菌属的丰度增加及有益菌属的丰度降低。结论 海参内脏可以改善溃疡性结肠炎小鼠的结肠组织病理表型、减少肠道出血、降低血清促炎因子和增加血清抗炎因子水平、缓解肠道微生物的失调,治疗溃疡性结肠炎作用显著。 |

Objective To study the therapeutic effect of holothurian visceral extract on ulcerative colitis in mice. Methods In this study, by exploring the effects of different concentrations of holothurian visceral extract on the disease activity index (DAI), colonic histopathology, serum inflammatory factors, and intestinal microorganisms in mice with ulcerative colitis induced by dextran sulfate sodium (DSS), its therapeutic effect on ulcerative colitis was evaluated. Results Both ethanol extract (EEx) and water extract (WEx) of holothurian viscera can dose-dependently alleviate the weight loss and increased DAI score caused by DSS in mice, alleviate the shortening of the colon in mice, accelerate the recovery of colonic tissue damage, reduce the content of fecal hemoglobin, and reduce the levels of pro-inflammatory factors nitric oxide (NO), tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and IL-6 in serum, and increase the level of the anti-inflammatory factor IL-10. At the same time, EEx and WEx can reduce the abundance of harmful genera such as Blautia, Helicobacter, and Erysipelatoclostridium in intestinal microorganisms, and increase the abundance of beneficial genera such as norank_f__Muribaculaceae and Lachnospiraceae_NK4A136_group, and alleviate the increase in the abundance of harmful genera and the decrease in the abundance of beneficial genera caused by DSS. Conclusion Holothurian viscera can improve the colonic histopathological phenotype of mice with ulcerative colitis, reduce intestinal bleeding, reduce serum pro-inflammatory factors, and increase serum anti-inflammatory factor levels, and alleviate the imbalance of intestinal microorganisms, and has a significant therapeutic effect on ulcerative colitis. |
