      目的 挖掘深海冷泉青霉中具有生物活性的次级代谢产物,丰富冷泉微生物次级代谢产物结构类型。方法 使用大米固体培养基对Penicillium javanicum OUCF108进行大发酵,积累次级代谢产物。采用有机溶剂萃取,正、反相硅胶柱层析将发酵产物初步分离,随后利用琼脂扩散法筛选活性组分并利用半制备高效液相对单体化合物进行分离纯化,通过质谱(MS)、核磁共振(NMR)数据分析以及文献比对,确定化合物结构,同时对所有化合物进行抗菌以及细胞毒活性测试。结果 从深海冷泉来源P. javanicum OUCF108中分离得到了3个蒽醌类化合物,7-chloroemodin(1),endocrocin(2),2-chloroemodic acid(3),2个二酮哌嗪类化合物,cyclo (6a'- (α, α-dimethylallyl)-L-Trp-L-Trp)(4),cyclo (8a-(α, α-dimethylallyl)-L-Trp-8a- (α, α-dimethylallyl)-L-Trp)(5)以及1个丁内酯类化合物(R)-striatisporolide A(6),其中化合物1对多重耐药菌Staphylococcus aureus CCARM 3090以及Enterococcus faeclis CCARM 5172展示出中等的抑菌活性,化合物5对K-562显示出细胞毒活性。结论 以活性为导向,从深海冷泉来源P. javanicum OUCF108中挖掘出带有潜在药用价值的蒽醌类及二酮哌嗪类化合物。
English Summary:
      Objective To explore bioactive secondary metabolites from deep-sea cold seeps-derived P. javanicum OUCF108, enriching their structural diversity. Methods P. javanicum OUCF108 was fermented on rice solid medium in a large-scale . The resulting fermentation products were separated by organic solvent extraction, normal-phase and reverse-phase silica gel column chromatography, and semi-preparative high performance liquid chromatography (HPLC). The structures of the compounds were determined by the analysis of MS and NMR data, along with comparison to the literatures. All the compounds were tested for antibacterial activity and cytotoxic activity. Results Three anthraquinone compounds, 7-chloroemodin (1), endocrocin (2) and 2-chloroemodic acid (3), two dione piperazine compounds, cyclo (6a'- (α, α-dimethylallyl)-L-Trp-L-Trp) (4) and cyclo (8a-(α, α-dimethylallyl)-L-Trp-8a- (α, α-dimethylallyl)-L-Trp) (5), and one butenolide compound, (R)-striatisporolide A(6) were isolated from the fermentation products of P. javanicum OUCF108. Compound 1 exhibited moderate antibacterial activity against multidrug-resistant strains Staphylococcus aureus CCARM 3090 and Enterococcus faecalis CCARM 5172. Compound 5 showed cytotoxic activity against K-562 cells. Conclusion Guided by antibacterial activities, anthraquinone and dione piperazine compounds were isolated from the fermentation products of P. javanicum OUCF108, which may be of potential medicinal value.
