      目的 研究1株中国西沙群岛的海绵共附生真菌Aspergillus aculeatus(15XS39ZM-1)的次级代谢产物。方法 利用薄层色谱法(TLC)、硅胶柱层析、Sephadex LH-20凝胶柱层析、中压制备液相色谱法(MPLC)、高效液相色谱法(HPLC)等方法,对菌株次级代谢物进行分离纯化;通过核磁共振波谱(NMR)及质谱(MS)等现代波谱学方法,结合相关文献数据分析,鉴定化合物结构。结果 从真菌Aspergillus aculeatus(15XS39ZM-1)代谢产物中分离鉴定了5个化合物,分别为29-N-demethylparaherquamide J (1)、29-N-demethylparaherquamide K (2)、paraherquamide J (3)、oxaline (4)、meleagrinb (5),其化学结构类型皆属于吲哚生物碱类化合物。斑马鱼抗炎活性测试结果表明,化合物3和4具有显著抗炎活性。抑制率分别为48.33%及37.57%。
English Summary:
      Objective To investigate the secondary metabolites of a sponge-associated fungus Aspergillus aculeatus (15XS39ZM-1) collected from the Xisha Islands of China were studied. Methods The secondary metabolites were isolated and purified by thin layer chromatography (TLC), silica gel column chromatography, Sephadex LH-20 gel column chromatography, medium pressure liquid chromatography (MPLC), high performance liquid chromatography (HPLC) by nuclear MR spectroscopy (NMR) and mass spectroscopy (MS) and data analysis. Results Five compounds were identified from the metabolites of fungus Aspergillus aculeatus (15XS39ZM-1), namely 29-N-demethylparaherquamide J (1), 29-N-demethylparaherquamide K (2), paraherquamide J (3), oxaline (4) and meleagrin (5). Their chemical structure types belong to indole alkaloids. The results of the anti-inflammatory activity test in zebrafish showed that compounds 3 and 4 had significant anti-inflammatory activity. The inhibition rates were 48.33% and 37.57%, respectively.
