      研究1株中国西沙群岛Plakortis galapagenis共附生真菌Aspergillus austroafricanus (18XS04ZP-2)的次级代谢产物。方法 利用薄层色谱法(TLC)、硅胶柱层析、Sephadex LH-20凝胶柱层析、中压制备液相色谱法(MPLC)、高效液相色谱法(HPLC)等方法,对菌株代谢物进行分离纯化;通过1H NMR、13C NMR及MS等现代波谱学方法,结合相关文献数据分析,鉴定化合物结构。结果 从真菌Aspergillus austroafricanus (18XS04ZP-2)代谢产物中分离得到10个化合物,分别为:4-hydroxy-17R-meth-ylinciste4rol(1),chaxinesB(2),volemolide (3),(22E,24R)-24-methylcholesta-7,22-dien-3β,5α,6β-triol(4), (22E,24R)-6β-methoxyergosta-5,22-diene-3β,5α-diol (5), (22E,24R)-3β,5α,9α-trihydroxy-ergosta-7,22-dien-6-one (6) , 6-dehydrocerevisterol (7),averufanin (8),versicolorin A (9),nidurufin (10)。活性测试结果,化合物2对K562细胞有一定的抑制作用,IC50为8.68μmol/L。
English Summary:
      To investigate the secondary metabolites and of the Aspergillus austroafricanus (18XS04ZP-2) isolated from a soft coral Plakortis galapagenis collected from the Xisha islands. Methods The secondary metabolites of the strain were separated and purified by thin layer chromatography(TLC),silica gel column chromatography,SephadexLH-20 gel column chromatography,medium pressure liquid chromatography(MPLC)and high performance liquid chromatography (HPLC). Through modern spectroscopy methods such as NMR and MS,combined with relevant literature data analysis to identify the structure of the compound. Results Ten compounds were isolated from Aspergillus austroafricanus (18XS04ZP-2), and they were identified as 4-hydroxy-17R-meth-ylincisterol (1), chaxines B (2),volemolide (3), (22E,24R)-24-methylcholesta-7,22-dien-3β,5α,6β-triol (4),(22E,24R)-6β-methoxyergosta-5,22-diene-3β,5α-diol (5), (22E,24R)-3β,5α,9α-trihydroxy-ergosta-7,22-dien-6-one (6) ,6-dehydrocerevisterol (7), averufanin(8),versicolorin A(9),nidurufin (10)。Activity test results,compound 2 exhibited inhibitory activity against K562, and the IC50 value was 8.68μmol/L.
