目的 研究1株药用红树老鼠簕(Acanthus ilicifolius L.)来源篮状菌属真菌Talaromyces flavus TGGP35的次级代谢产物及其抗氧化活性。方法 综合利用正相硅胶柱层析、Sephadex LH-20凝胶柱层析、半制备高效液相色谱(HPLC)等色谱分离方法,对TGGP35液体发酵产物的乙酸乙酯粗浸膏进行系统分离纯化;运用核磁共振波谱(1D NMR)、质谱(MS)等方法并结合文献报道数据确定化合物结构,并对其抗氧化活性进行测试。结果 从真菌T. flavus TGGP35发酵产物中分离得到9个化合物,包括2个生物碱类化合物thymidine(1)和2"-O-methyluridine(2),3个内酯类化合物aspergilactone A(3)、aspergilactone B(4)和nafuredin(5),1个糖苷类化合物carnemycin E(6),3个二酮哌嗪类化合物cyclo-(L-Val-L-Pro)(7)、cyclo(L-Pro-L-Leu)(8)和cyclo(L-Pro-L-Leu)(9),其中化合物6表现出一定的抗氧化抑制活性,其IC50值为0.37 mM。结论 化合物1~4和6为首次从篮状菌属真菌(Talaromyces sp.)中分离得到,发现1个具有中等抗氧化活性的化合物,具有一定的研究和应用价值。 |

Abstract: Objective To study the secondary metabolites and antioxidant activities of the fungus Talaromyces flavus TGGP35 derived from medicinal mangrove Acanthus ilicifolius L. Methods The isolated compounds were separated by normal phase silica gel, Sephadex LH-20, semi-preparative HPLC column chromatography, thin layer chromatography and so on. And their structures were identified by 1D NMR, MS, other spectral analysis methods and comparison with literature data. They were also tested for their antioxidant activity. Results Nine known compounds were isolated from the fungus T. flavus TGGP35, including two alkaloids thymidine (1) and 2"-O-methyluridine (2), three lactones aspergilactone A (3), aspergilactone B (4), nafuredin (5), one glycoside carnemycin E (6), and three diketopiperazines cyclo-(L-Val-L-Pro) (7), cyclo (L-Pro-L-Leu) (8) and cyclo (L-Pro-L-Leu) (9). Among them, compound 6 showed antioxidant inhibitory activity with IC50 value of 0.37 mM. Conclusion Compounds 1-4 and 6 were isolated from the genus Talaromyces sp. for the first time, and compound 6 showed moderate antioxidant activity, these results showed important research and application value. |
