目的 探讨纳豆芽孢杆菌发酵菲律宾蛤仔制备的生物活性肽(RBPs)对去卵巢大鼠(OVX)骨质疏松症的改善作用,为我国丰富的贝类资源的高值化利用提供依据。方法 用RBPs改善去卵巢骨质疏松症大鼠5周后,检测血清中碱性磷酸酶(ALP)、25-羟基维生素D(25(OH)D)的含量,利用Micro-CT、HE染色分析骨密度、骨微结构改变情况,并进一步探究RBPs对破骨和成骨通路的调控作用。结果 RBPs使OVX大鼠的血清ALP含量显著降低,25(OH)D含量升高,骨密度(BMD)、骨钙含量和骨微结构均有明显改善。在分子机制上,我们发现RBPs可以通过抑制RANKL/RANK/OPG信号通路减少骨吸收,并激活Wnt/β-catenin通路促进骨形成。结论 RBPs对绝经后的骨质疏松症具有良好的改善作用,为RBPs作为一种改善绝经后骨质疏松症的海洋药物来源提供了依据。 |

Objective To investigate the effect of bioactive peptides from Ruditapes philippinarum fermented with Bacillus natto (RBPs) in an ovariectomized rat (OVX) model, and to provide evidence for the high-valued utilization of abundant shellfish resources in China. Methods After 5 weeks of treatment with RBPs, the contents of alkaline phosphatase (ALP) and 25-hydroxyvitamin D (25(OH)D) in serum were detected. The changes of BMD and bone microstructure were analyzed by micro-CT and HE staining and the regulation of RBPs on osteoclast and osteogenesis pathways were explored. Results The content of ALP decreased significantly, and the content of 25(OH)D increased significantly. Meanwhile, BMD, bone calcium content and bone microstructure were significantly improved. In molecular mechanism, we found that RBPs can reduce bone resorption by inhibiting RANKL/RANK/OPG signaling pathway and activate the Wnt/β-catenin pathway, promoting bone formation. Conclusion RBPs can improve significantly postmenopausal osteoporosis, and can be a potential marine drug source for the treatment of menopausal osteoporosis. |
