目的 为优化提取仿刺参精活性肽并考察其对酒精性肝损伤的保护作用。方法 采用单因素和响应面试验优化活性肽酶解释放工艺参数,测定切向流超滤系统分级分离组分体外抗氧化活性,通过构建酒精性肝损伤动物模型,检测分析低聚肽对小鼠生化指标和肝脏病理组织学影响。结果 通过筛选优化得出碱性蛋白酶为水解仿刺参精的最优用酶,在酶解温度55.33 ℃、pH 9.44、酶解时间5.72 h、加酶量6 kU/g的优化条件下,水解度达到了45.40%;4个超滤组分均具有较好的清除DPPH、羟基和超氧阴离子自由基的活性,其中<1 kDa低聚肽作用最强;动物实验表明低聚肽可缓解酒精引起的体重下降,高剂量组(800 mg/kg)、中剂量组(400 mg/kg)动物血清中AST、CHOL、TBIL、LDL含量和肝细胞变性程度均显著低于模型动物(P<0.05),高剂量组(800 mg/kg)SOD水平显著高于模型动物(P<0.05)。结论 仿刺参精低聚肽可以提高机体的抗氧化能力,对酒精性肝损伤具有明显的保护作用。通过本研究以期为酒精性肝损伤预防或治疗研究提供理论和应用基础。 |

To optimize the extraction of active peptide from Apostichopus japonicus sperm and investigate its protective effect on alcoholic liver injury. Methods Single factor and response surface methodology were used to optimize the enzymatic hydrolysis process parameters. Antioxidant activity in vitro of the fractionated components by tangential flow ultrafiltration system was determined. Finally, by constructing an animal model of alcoholic liver disease, the effects of oligopeptides on biochemical indexes and liver histopathology in mice were detected and analyzed. Results The results showed that alkaline protease was the best enzyme for hydrolyzing Apostichopus japonicus sperm. Under the optimized conditions of enzymatic hydrolysis temperature 55.33 ℃, pH 9.44, time 5.72 h, and enzyme dosage 6 kU/g, the degree of hydrolysis reached 45.40%; All the four ultrafiltration components had good scavenging activities of DPPH, hydroxyl and superoxide anion radicals, and the oligopeptide <1 kDa had the strongest scavenging activity; Oligopeptides could alleviate the weight loss of animals caused by alcohol, and the levels of AST, CHOL, TBIL, LDL and degree of hepatocellular degeneration of high dose (800 mg/kg) and medium dose (400 mg/kg) groups were significantly lower than those in model animals (P<0.05).The SOD level in high dose group (800 mg/kg) was significantly higher than that in model animals (P<0.05). Conclusion It is concluded that the oligopeptides of Apostichopus japonicus sperm can improve the body"s antioxidant capacity and have obvious protective effects on alcoholic liver disease. Through this research, we hope to provide theoretical and application basis for the prevention or treatment of alcoholic liver injury. |
