      摘 要:目的 研究1株红树植物木果楝(Xylocarpus granatum Koenig)来源内生真菌Phomopsis sp. MGGP14112的次级代谢产物及其生物活性。方法 综合运用硅胶柱层析、Sephadex LH-20凝胶柱层析、半制备高效液相色谱(HPLC)等色谱分离技术对菌株的发酵产物进行分离纯化,通过核磁共振波谱(NMR)、质谱(MS)等多种现代波谱技术以及与文献数据对照,确定化合物的结构,对化合物进行抗氧化活性和抗虫活性测试。结果 从内生真菌Phomopsis sp.中分离鉴定出7个化合物1~7,结构分别为phomoxydiene B (1)、deacetylmycoepoxydiene (2)、mycoepoxydiene (3)、5,6-dihydroxy-2,3,6-trimethylcyclohex-2-enone (4)、4-methoxy-3,6-dimethylpyran-2-one (5)、petasol (6)和lithocarin A (7)。其中化合物3表现出一定的抗氧化活性,其IC50为3.83 mg/mL;化合物1~3和7对棉铃虫幼虫(Helicoverpa armigera Hubner)具有一定的生长抑制活性,其IC50值为200 μg/mL。结论 从红树内生真菌Phomopsis sp. MGGP14112中分离获得7个单体化合物,活性筛选发现1个具有抗氧化活性和4个具有棉铃虫幼虫生长抑制活性的化合物,具有重要的研究和应用价值。
English Summary:
      Abstract: Objective To investigate the bioactive secondary metabolites from the fermentation of mangrove Xylocarpus granatum Koenig-derived fungus Phomopsis sp. MGGP14112. Methods The secondary metabolites were isolated and purified by column chromatography silica gel, Sephadex LH-20 and semi-preparative HPLC. Their structures were identified by 1D NMR, MS, other spectral analysis methods and comparison with literature data. The antioxidant and insecticidal activities were also tested. Results Seven known compounds were isolated from the endophytic fungus Phomopsis sp. MGGP14112, including phomoxydiene B (1)、deacetylmycoepoxydiene (2)、mycoepoxydiene (3)、5,6-dihydroxy-2,3,6-trimethylcyclohex-2- enone (4)、4-methoxy-3,6-dimethylpyran-2-one (5)、petasol (6) and lithocarin A (7). Compound 3 exhibited potent antioxidant activity with the IC50 values of 3.83 mg/mL. Compounds 1~3 and 7 showed potent inhibitory activity against Helicoverpa armigera Hubner with the same IC50 values of 200 μg/mL. Conclusion Seven compounds were isolated from the fungus Phomopsis sp. MGGP14112. One compound showed potent antioxidant activity, and four compounds showed potent inhibitory activity against Helicoverpa armigera Hubner, which has important research and potential development value.
