      目的 探究海藻提取物对乙醇诱导小鼠急性胃溃疡的保护作用及其作用机制。方法 将8周龄雌性健康BABL/C小鼠随机分为正常组、模型组、海藻提取物低剂量组(7.2 mg·kg-1·bw-1)、海藻提取物高剂量组(21.5 mg·kg-1·bw-1)。预灌胃2周后,模型组和海藻提取物组灌胃10 mL·kg-1·bw-1 85%乙醇,建立小鼠急性胃溃疡模型。测定各组小鼠胃腺部出血状况、胃溃疡抑制率,以及血清谷草转氨酶(Aspartate aminotransferase,AST)、谷丙转氨酶(Alanine aminotransferase,ALT),丙二醛(Malondialdehyde,MDA)、超氧化物歧化酶(Superoxide Dismutase,SOD)和白介素-6(Interleukin-6,IL-6)、肿瘤坏死因子-α(Tumor?necrosis?factor-α,TNF-α)含量,进一步检测胃组织中核转录因子κB(Nuclear?factor?κB,NF-κB P65)、pNF-κB P65的蛋白表达水平。结果 海藻提取物可显著改善模型小鼠胃溃疡病理学变化,且呈剂量效应关系提高SOD的表达水平,降低MDA、AST、ALT、TNF-α和IL-6表达水平,以及NF-κB通路关键蛋白(IkB、p-IkB、NF-κB P65、pNF-κB P65)的表达。结论 海藻提取物通过调节NF-κB介导的炎症通路,对乙醇所致急性胃溃疡有保护作用。
English Summary:
      Objective In this study, the protective efficacy and mechanism of seaweed extracts on acute gastric ulcer mice induced by ethanol were investigated in vivo. Methods 8-week-old female healthy BABL/c mice were randomly allocated into Normal group, Model group, low-dose of seaweed extracts group (7.2 mg·kg-1·bw-1), high-dose of seaweed extracts group (21.5 mg·kg-1·bw-1). After 2 weeks intragastric?administration, the group of model and seaweed extracts were given by?gavage for 10 ml·kg-1·bw-1 85% ethanol to establish the acute gastric ulcer model. Then the level of ulcer bleeding, ulcer incidence rate, aspartate aminotransferase (AST), alanine aminotransferase (ALT), malondialdehyde (MDA), superoxide dismutase (SOD), interleukin-6 (IL-6), tumor?necrosis?factor-α (TNF-α) were measured, and protein expression of inflammation factor (Nuclear?factor-κB P65, pNF-κB P65) in?stomach?tissue of mice were assayed with western blot. Results Intragastric administration of seaweed extracts significantly lowered the ulcer index value and ulcer incidence rate compared to the ulcer control group, increased SOD levels, decreased MDA, AST, ALT, TNF-α, IL-6 levels and the protein expression levels of NF-κ B pathway (IkB, p-IkB, NF-κB p65, pNF-κB p65). Conclusion Seaweed extracts has a gastric protective activity against ethanol on ethanol and this might be related to regulate NF-κB-mediated inflammatory pathway.
