目的 制备水溶性可得然胶低聚糖,并研究其对RAW264.7细胞的激活作用。方法 以商业化生产的可得然胶为原料,采用多种降解方式进行水溶性可得然胶低聚糖制备工艺的探索,对分子量、总糖含量和水溶性等理化性质进行考察,确定最佳降解条件制备水溶性可得然胶并进行分离纯化获取不同分子量的低聚糖组分;测定了水溶性可得然胶低聚糖对RAW264.7细胞增殖、吞噬能力和细胞因子(TNF-α和IL-6)释放的影响。结果 以组合降解的方式制备得到了3种不同分子量水溶性可得然胶低聚糖组分。分子量为2.01 kDa的水溶性可得然胶低聚糖CDOS-3可以显著促进RAW264.7细胞的增殖和对中性红的吞噬,而分子量为22.32 kDa的水溶性可得然胶低聚糖CDOS-1则可以显著提高细胞因子的释放。结论 不同分子量的水溶性可得然胶低聚糖能够激活RAW264.7细胞,是1种潜在的免疫应答调节剂。 |

Objective To investigate the immunostimulatory effect of soluble low-molecular-weight curdlan on RAW264.7 cell. Methods The commercial curdlan was used as raw material, and the preparation of soluble low-molecular-weight curdlan (CDOS) was explored by different degradation methods. The molecular weight, sugar content and water-solubility of the degradation products were investigated to determine the best degradation conditions for the preparation of soluble curdlan. The different molecular weight CDOS were obtained by separation and purification. In the immunostimulatory assay, cells proliferation, phagocytosis, cytokines (TNF-α and IL-1β) release were determined to study the effects of CDOS on RAW264.7 cell. Results CDOS-3(Mw: 2.01 kDa) and CDOS-1(Mw: 22.32 kDa) could significantly improve the cell proliferation, the neutral red phagocytosis and cytokines release of RAW264.7 cell, respectively. Conclusion CDOS could virtually activate RAW264.7 cell via multiple levels and be a potential immune response regulator. |
