目的 从海洋放线菌中筛选出对白色念珠菌有抗菌活性的菌株,预期获得新结构活性物。通过优化活性菌株的发酵条件,以期提高活性菌株产生活性物产量。方法 本研究以白色念珠菌作为靶标,采用抑菌圈法,从实验室保存的2632个放线菌株中筛选出对白色念珠菌有抗菌活性的菌株,并优化菌株的发酵条件。结果 筛选出对白色念珠菌有较强抑制作用的海洋放线菌H74-18菌株,明确了它对作物疫病菌和炭疽病菌有抑制作用活性,其优化发酵条件为:培养基为摇瓶培养基,培养温度为28℃,接种龄为48h,接种量为10%,摇床转速为300 r/min,发酵时间为72h。结论 菌株H74-18对白色念珠菌抗菌活性较高且抗菌谱较广。通过优化发酵条件可以提高其活性物产量,说明它具有进一步研究开发的应用前景。 |

Objective It was to screen out the strains with inhibitory activity against Candida albicans from marine actinomycetes and expect to obtain new structural active compounds. By optimizing the fermentation conditions of the active strains, it was hoped to improve the production of the active strains. Methods Candida albicans was used as the target. From 2632 actinomycetes preserved in the laboratory, the strains with inhibitory activity against Candida albicans were screened. Then the fermentation conditions of the active strain were optimized. Results The strain H74-18 with strong inhibitory effect on Candida albicans was screened out. The inhibition activity against Phytophthora and Colletotrichum was determined. The optimal fermentation conditions of strain H74-18 were as follows: the culture medium was the shake flask culture, the culture temperature was 28 ℃, the inoculation age was 48h, the inoculation amount was 10%, the rotating speed of shaking table was 300r / min, and the fermentation time was 72h. Conclusion The inhibitory activity of H74-18 against Candida albicans was high and its spectrum against pathogen was wide. By optimizing the fermentation conditions, the yield of its active substance could be increased. It shows that it has the application prospect of further research and development. |
