      摘 要:目的 验证一种新型补钙剂--焦谷氨酸钙(calcium L-pyroglutamate, CAP)的抗疲劳作用。方法 选取正常雄性昆明小鼠作为实验动物,腹腔注射脂多糖形成炎症模型,随机分组并通过观察小鼠负重游泳时间,血清尿素氮(BUN)、乳酸(LA)水平,血清乳酸脱氢酶(LDH)活性,血清肿瘤坏死因子α(TNF-α),白介素10(IL-10)、白介素1β(IL-1β),肝糖原、肌糖原以及肌肉组织中丙二醛(MDA)含量,超氧化物歧化酶(SOD)活性,综合评价焦谷氨酸钙的抗疲劳作用。结果 与模型组相比,高、低剂量组小鼠负重游泳时间显著延长(P<0.05或P<0.01),焦谷氨酸钙高剂量组BUN水平显著下降(P<0.01),血清LA水平显著下降,血清LDH活性显著提升,血清TNF-α,IL-1β浓度显著降低,IL-10浓度显著提高,肝、肌糖原含量显著升高,肌肉SOD活性显著升高,MDA含量显著下降。结论 焦谷氨酸钙具有显著抗疲劳效果,其作用机制可能与调节能量代谢,抗氧化应激活性以及抗炎症活性有关,可以作为一种潜在的抗疲劳药物。
English Summary:
      Abstract:Objective To study the anti-fatigue effect of calcium L-pyroglutamate (CAP, a new type of calcium supplement) in healthy mice. Method Healthy male Kunming mice were chosen as experimental animals and randomly assigned to several groups, orally administrated with correlated medicine once a day for 28 days. The weight-loaded forced swimming experiment of mice was performed to record the swimming time of mice. The content of blood urea nitrogen (BUN), serum lactic acid (LA) as well as serum lactic dehydrogenase (LDH) activity after a 30 min swimming were measured, meanwhile, the hepatic glycogen and muscle glycogen as well as inflammatory factors ( including TNF-α, IL-1β and IL-10), oxidant stress related indexes including superoxide dismutase (SOD) and malonaldehyde (MDA) were detected to study the anti-fatigue effect of CAP in mice. Results The results showed that calcium L-pyroglutamate could significantly prolong the time of weight-loading swimming, compared with the model group (P<0.01 or P<0.05). The content of BUN and lactic acid in serum was significantly decreased in the high-dose group (P<0.05), the activity of LDH was significantly increased in positive control group and the high-dose group as well as the low-dose group (P<0.01 or P<0.05). Calcium L-pyroglutamate increased the levels of glycogen in both liver and muscle, serum concentrations of TNF-α, IL-β as well as muscle MDA were significantly decreased, while serum IL-10 concentration and SOD activity were significantly increased. Conclusion Our results suggested that calcium L-pyroglutamate has anti-fatigue effect in healthy mice which might be related to its regulation effect on energy metabolism and anti-oxidant activity as well as inflammatory factors, and thus could be a potential functional factor in anti-fatigue functional foods.
