      目的 对1株分离自福建沿海的橘绿木霉Trichoderma citrinoviride NM-035所发酵产生的抗肿瘤活性成分进行研究。方法 将纯化得到的菌株在28摄氏度下静置培养30天,发酵液经乙酸乙酯萃取得到总浸膏;先后利用硅胶柱层析、凝胶柱层析以及半制备型高效液相等色谱手段,对总浸膏进行分离纯化;通过核磁共振和高分辨电喷雾质谱方法并结合文献,对产物的结构进行解析并最终对其活性进行初步评价。结果 从该菌株中分离得到了1个新的tetramic acid型生物碱,并命名为penicillenol F (1)。该化合物对人恶性黑色素瘤和肝癌细胞具有一定的抑制活性。 结论 从橘绿木霉Trichoderma citrinoviride NM-035中发现1个新的具有抑制人恶性黑色素瘤和肝癌细胞活性的生物碱类化合物penicillenol F(1)。
English Summary:
      Objiective To study the antitumor chemical constituent of a marine-derived fungus Trichoderma citrinoviride NM-035 collected from Fujian coast. Methods The purified strain was static incubated at 28 ℃ for 30 days. The broth was extracted by ethyl acetate to obtain the crude extract, which was successively purified by silica gel, Sephadex LH-20 and semi-preparative HPLC. The structure of purified compound was identified by NMR and MS analysis as well as comparison with literatures, and its antitumor activity was evaluated. Results A novel alkaloid with a tetramic acid skeleton, named penicillenol F (1) was isolated from a marine-derived fungus Trichoderma citrinoviride, which showed weak cytotoxic effects against the cell lines of malignant melanoma and liver cancer. Conclusion A novel alkaloid, penicillenol F (1) with cytotoxic activity was isolated from Trichoderma citrinoviride.
