      摘要:目的 从马尾藻中提取活性多肽,并研究其在抗菌和抑制血栓形成方面的活性。 方法sephdexG-50凝胶层析柱和高效液相色谱柱分离纯化多肽组份,以280nm监测样品;Molish实验检测提取的多肽含糖量;用比浊法检测不同剂量的马尾藻多肽(0、10、20、40、60、80 μg/mL)对大肠埃希菌(D1314)和金黄色葡萄球菌(s.agr+ 、RN4220)生长的影响;将大鼠随机分为生理盐水组、马尾藻多肽(100、200、400μg/mL)、肝素1600U/mL组,分别给予相应药物,电刺激大鼠颈动脉形成血栓,观察大鼠颈动脉血栓形成的情况。 结果 可从马尾藻匀浆上清液中获得2个收集峰,主要多肽集中在B收集峰;高效液相分析分析B收集峰由6个组份组成;Molish实验显示多肽组份不含多糖类物质;比浊法显示多肽组份具有抑制大肠埃希菌的生长,而对金黄色葡萄球菌不敏感;且通过电刺激大鼠颈动脉血栓形成,发现该多肽具有抑制血栓形成的作用。 结论 我们成功从马尾藻中提取活性多肽组份,该多肽组份具有抑制大肠埃希菌生长和血栓形成的作用。
English Summary:
      Abstract: Objective The studied polypeptides activities in antibacterial and antithrombotic activities were extracted from sargassum. Method The polypeptide components were isolated and purified by sephdexg-50 gel chromatography and HPLC, and the samples were monitored at 280 nm. Molish experiment detected the sugar content of polypeptide. Effects of different doses of sargassum polypeptide (0, 10, 20, 40, 60, 80 g/mL) on growth of Escherichia coli (D1314) and Staphylococcus aureus (s.agr+, RN4220) were detected by using the method of turbidity. Rats were randomly divided into normal saline group, polypeptides (100, 200, 400 g/mL) and heparin (1600U/mL) groups, and given corresponding drugs, respectively, to electrically stimulate the carotid thrombosis , and observe the situation of carotid thrombosis . Results Two collection peaks can be obtained from the supernatant of sargassum, the main polypeptides were concentrated in the B collecting peak.The analysis of B collection peak consists of 6 components by HPLC. Molish experiments showed that the polypeptide components did not contain polysaccharides. The polypeptide can inhibit the growth of Escherichia coli, and is not sensitive to Staphylococcus aureus. By electrical stimulation of carotid thrombosis in rats, it was found that the polypeptide could inhibit thrombosis. Conclusion We have successfully extracted the active polypeptide from Sargassum , which can inhibit the growth and thrombosis of Escherichia coli.
