摘要:目的 研究浒苔多糖EP2对小鼠免疫功能的影响。方法 将Babl/c小鼠随机分成6组,1~3组为正常小鼠,4~6组为CTX诱导的免疫抑制小鼠,各组每日分别以0、50、125、0、50、125 mg/kg的剂量灌服EP2,持续14 d。按常规方法测定小鼠血常规指标、器官指数和巨噬细胞吞噬功能,采用耳肿法测定迟发型变态反应(DTH),采用MTT法测定脾脏淋巴细胞增殖活性,采用流式细胞术分析T淋巴细胞亚群,采用酶联免疫吸附试验检测细胞因子和BSA抗体。 结果 给予EP2的免疫抑制小鼠脾脏和胸腺指数、血液WBC、PMBC、LY、RC、CD8+细胞含量、淋巴细胞增殖活性、血清IL-2、IL-4、IFN-γ和BSA抗体水平显著升高;巨噬细胞吞噬功能、DTH显著增强;CD4+/CD8+比例显著下降。给予高剂量EP2的正常小鼠脾脏和胸腺指数、WBC、PMBC、RC含量、淋巴细胞增殖活性、CD4+、CD8+和CD4+/CD8+比例、血清IL-2及BSA抗体水平显著升高;巨噬细胞吞噬功能显著提高;DTH、血清IL-4和IFN-γ水平呈升高趋势。结论 EP2能显著增强正常小鼠和免疫抑制小鼠的免疫功能,且呈现明显量效关系 |

Abstract:Objective The present work was aimed to inverstigate the effects of EP2 isolated from Enteromorpha on immune function of mice. Methods The Babl/c mice were randomly divided into 6 groups, mice in group 1~3 were normal and mice in group 4~6 were pre-treated by cyclophosphamide to induce immunosuppression. The mice above were respectively gavaged 0, 50, 125, 0, 50 and 125 mg/kgw EP2 per day for 14 days. The blood cell content, indexes of immune organs, phagocytic function of macrophage were analyzed by regular method, the degree of delayed type hypersensitivity(DTH) of the mice were evaluated by ear sweelling assay, the proliferation of T and B lymphocytes was measured by MTT assay, the T cell subsets were analysed by flow cytometry, the serum cytokines and BSA antibody titers were determined by ELISA. Results The index of spleen and thymus, the content of white blood cells(WBC), peripheral blood mononuclear cells(PMBC), lymphocytes(LY), reticulocytes(RC) and CD8+, the proliferation of T and B lymphocytes and serum content of IL-2、IL-4、IFN-γ and BSA antibody titers of the immunosuppressed mice administered EP2 were significantly increased, the phagocytic function of macrophages and the DTH were also elevated significantly, and the evident decreased ratio of CD4+/CD8+ were simultaneously observed in those immunosuppressed mice received EP2. As for normal mice, the index of spleen and thymus, the content of WBC, PMBC, LY, RC, CD4+, CD8+, the ration of CD4+/CD8+,the proliferation of T and B lymphocytes, the phagocytic function of macrophages, the serum IL-2 and BSA antibody levels were remarkably improved owing to high-dose EP2 supplementation. Moreover, the DTH, the serum IL-4 and IFN-γ content tend to be higher than that of control. Conclusion The EP2 fraction of Enteromorpha polysaccharids can be used to improve immune function of the normal mice and immunosuppressed mice in a dose-dependent manner. |
