摘 要:目的:研究红纤维虾海藻黄酮的大孔树脂纯化工艺及抗氧化活性。方法:通过静态吸附和解析实验,从5种大孔树脂中筛选出纯化黄酮效果最优的树脂,并确定该树脂的最佳工艺条件。以Vc为对照品,考察红纤维虾海藻黄酮对DPPH自由基(DPPH?)和羟基自由基(?OH)的抗氧化活性。结果:HPD600纯化红纤维虾海藻黄酮效果最好,最佳纯化工艺为:上样液pH 3~4,黄酮浓度1.0 mg/mL,以1.5 mL/min的流速上样16 BV,洗脱剂为75%乙醇,以2.0 mL/min的流速洗脱,洗脱体积为8 BV,在此条件下黄酮纯度可达76.39%。红纤维虾海藻黄酮对DPPH?和?OH有良好的清除作用。结论:HPD600大孔树脂对红纤维虾海藻黄酮纯化效果良好,可用于红纤维虾海藻黄酮的纯化,且纯化后的红纤维虾海藻黄酮具有较强的抗氧化活性。 |

Abstract: Objective: To study the flavonoids purification process by macroporous resin and antioxidant activity of flavonoids from Phyllospadix iwatensis Makino. Methods: Five kinds of macroporous resin were selected to compare their purification ability by dynamic adsorption and desorption capacities as the indicators and the best one was used to optimize the purification process. The antioxidant activity of the flavonoids from Phyllospadix iwatensis Makino was tested by scavenging DPPH? and ?OH with Vc as references. Results: HPD600 was the most suitable type of macroporous resin to purify flavonoids from Phyllospadix iwatensis Makino and the optimum conditions were as follows: the sample pH was 3~4,the flavonoids concentration was 1.0 mg/mL, loading 16 BV at a ?ow rate of 1.5 mL/min and subsequently eluting the column with 8 BV of 75% ethanol at a ?ow rate of 2.0 mL/min, the purity of flavonoids could reach 76.39% in this condition. The flavonoids from Phyllospadix iwatensis Makino had good effect to scavenge DPPH? and ?OH. Conclusion: HPD600 macroporous resin could be used for purification of flavonoids from Phyllospadix iwatensis Makino with good purification capability, and the flavonoids after purification had strong antioxidant activity. |
