利用响应面法对薄膜分散法制备鱼油纳米脂质体的处方进行优化,初步测定了其理化性质。并研究了用南极磷虾卵磷脂制备鱼油纳米脂质体的影响因素,主要包括磷脂浓度、胆固醇含量、吐温-80含量和鱼油含量,并通过响应面法优化制备工艺。最终得到的最佳处方为:磷脂浓度25 mg/mL,m(磷脂):m(胆固醇)=2.5:1,m(磷脂):m(鱼油)=6.5:1,吐温-80含量为17%。此时鱼油纳米脂质体的粒径为21.20 nm,包封率为88.24%,zeta电位为-8.34 mV,接近于模型预测值。 |

Response surface methodology(RSM) was selected to optimize the fish oil nanoliposomes prescription by thin film hydration(TFH), determinated the initial physical and chemical properties.And research in the factors that influence the fish oil nanoliposomes with the phosphatidylcholine from Antarctic krill.Mainly includes the content of phosphatidylcholine ,cholesterol ,Tween-80 and fish oil,then optimized the preparation through the RSM.Finally we obtain the best prescription was as follows:phosphatidylcholine content was 25 mg/mL,the ratio of phosphatidylcholine and cholesterol was 2.5:1,the ratio of phosphatidylcholine and fish oil was6.5:1,the content of Tween-80 was 17%. Under these conditions,mean diameter and polydispersity index of fish oil nanoliposomes were 21.20 nm and encapsulation efficiency(EE) was 88.24%,zeta potential was -8.38 mV,which was well matched with the predictive preparation. |
