      目的 探讨巨细胞病毒诱导的β-分泌酶在小鼠体内的变化及在藻酸双酯钠(PSS)治疗后淀粉样蛋白前体(APP)和β-分泌酶(BACE-1)的表达。方法 将30只雄性小鼠(雄性和雌性)随机分为3组(每组10只)。对照组小鼠不做任何处理;CMV感染组,腹腔一次性注射MCMV 0.2 mL,30 d后处死;PSS治疗组,CMV感染后,每日灌胃PSS 60 mg/kg,持续14至30 d。在第30天,处死所有的小鼠。通过蛋白质免疫印迹和免疫组化检测β-分泌酶的表达。结果 APP和BACE-1在感染小鼠脑中的表达显着高于PSS处理后,与感染组相比蛋白表达降低。结论 PSS可通过降低断奶小鼠巨细胞病毒诱导的β-分泌酶的表达水平,缓解继发性脑损伤程度,并在未来的治疗中起到保护神经作用。
English Summary:
      Objective To investigate the changes of β-secretase (BACE-1) induced by cytomegalovirus (CMV) in mice and the expression of amyloid precursor protein (APP) and β-secretase in mice treated with polysaccharide sulfate (PSS). Methods Thirty mice (male and female) were randomly divided into 3 groups (10 mice/group). In the negative group, mice were fed without any treatment; in the CMV infection group, mice received a single intraperitoneal injection of 0.2 ml murine CMV; in the PSS treatment group, after CMV infection, PSS was administered intragastrically at 60 mg/kg daily for two weeks. All the mice were sacrificed on day 30. The expression of BACE-1 was assessed by western blot and immunohistochemistry. Results The expression of APP and BACE-1 in the brain of infected mice was significantly higher than normal, but after PSS treatment, the protein levels decreased. Conclusions PSS relieves the degree of secondary brain injury by reducing the expression level of β-secretase induced by CMV in weaning mice and plays a neuro-protective role that might be exploited in future treatment.
