摘 要:目的 渤海海泥来源放线菌Streptomyces sp. LXF-80可以生产罕见的包含α-吡喃酮的三环骈合聚酮化合物肠道菌素(Enterocin)。美替拉酮是一种细胞色素P450氧化酶的抑制剂,本实验试图利用美替拉酮影响肠道菌素的生合成过程,表达获得新的聚酮类化合物。方法 向LXF-80的发酵培养基中添加酶抑制剂,结合HPLC分析确定最优条件,并进行大规模发酵和目标产物的分离。结果 从添加美替拉酮的LXF-80发酵提取物中共分离获得3个化合物,包括Enterocin(1)、5-deoxyenterocin(2)以及3-epi-5-deoxyenterocin(3)。结论 实验结果证明美替拉酮的添加,影响了菌株聚酮类化合物的生合成过程,得到两个肠道菌素的形成过程中的中间体化合物2和3。 |

Abstract:Objective Our recent investigation of marine-derived strain Streptomyces sp.LXF-80 resulted in the isolation of complicated tricyclicpolyketide enterocin. Metyrapone is a kind of cytochrome P450 enzyme inhibitor. In this paper, we try to detect novel polyketide compounds by affecting the biosynthesis pathways of the strain, based on the treatment with metyrapone. Methods We fed various enzyme inhibitors into the medium of Streptomyces sp.LXF-80, determined the optimal conditions according to the HPLC analysis and separated aim compounds. Results Enterocin(1), its two derivatives 5-deoxyenterocin(2) and 3-epi-5-deoxyenterocin(3) were separated. Conclusion Metyrapone could actually affect the metabolic pathways of Streptomyces sp. LXF-80. |
