      摘 要:目的 建立中药海藻中总甾醇的含量测定方法。方法 采用Bligh-Dyer方法提取总脂,以岩藻甾醇为对照品,磷硫铁试剂为显色剂,利用分光光度法进行含量测定,并进行方法学验证。结果 含量测定方法学结果表明,在检测波长576 nm下,岩藻甾醇在0.01 ~ 0.07 g ? L–1具有良好的线性关系,r = 0.9970;显色产物在90 min内稳定,RSD 0.54%;平均加样回收率为99.16%,RSD 1.43%(n = 6);方法重复性良好,RSD 1.05%(n = 6)。结论 建立的磷硫铁-分光光度法可用于中药海藻中总甾醇的含量测定。
English Summary:
      Abstract: Objective To establish a method for determination of total phytosterols in marine TCM Sargassum. Methods The total lipid was extracted by the Bligh-Dyer method. The spectrophotometric method was developed and validated using fucosterol as reference substance and phosphorus sulfur iron solvent as chromogenic reagent. Results The detection wavelength was set at 576 nm. The calibration curve of fucosterol and absorption spectrum was linear in the concentration range of 0.01 g ? L–1 ~ 0.07 g ? L–1 (r = 0.9970). The chromogenic product could keep stable within 90 minutes and the RSD was 0.54%. In addition, the repeatability (RSD 1.05%, n = 6) and average recovery rate 99.16% (RSD 1.43%, n = 6) met the determination requirements well. Conclusion The developed method could be used for quantitative determination of total phytosterols in marine TCM Sargassum.
