目的以响应面法优化梅花状青霉代谢抗氧化活性物质的培养条件。方法以目标菌株发酵液浸膏抗氧化活性为响应值,考察培养时间、碳源、氮源与浸膏抗氧化活性之间的相关性。以单因素实验结果为基础,利用Box-Behnken设计组合实验方案,采用Design-Expert 8.05 软件优化菌株的实验室培养条件参数。结果菌株代谢抗氧化活性物质最优培养条件为:培养时间为16 d,碳源为葡萄糖20.0 g·L?1,氮源为蛋白胨10.8 g·L?1,平行测试DPPH自由基清除率为97.83 %,其IC50为13.57 μg·mL?1。结论响应曲面法回归模型的预测值与实测值接近,拟合较好,该方法可用于类似菌株活性物质获取条件的优化。 |

Objective To optimize the antioxidant secondary metabolite processing of Penicillium herquei LGT201107. Methods The Radical Scavenging Activity (RSA) of the extracts from P. herquei was taken as the response value. The relativity between the extracts bioactivity and the incubation time, dosage of glucose and peptone was studied. The experimental methods were suggested following the Box-Behnken method and the single-factor experimental results. The labortary cunlture condition was optimized by the software of Design-Expert 8.05. Results The optimum culture condition was suggested as: incubation time 16 d, the dosage of glucose 20.0 g·L?1 and peptone 10.8 g·L?1, respectively. Within optimized condition, the RSA of extracts from P. herquei was up to 97.83 %, and the IC50 was 13.57 μg·mL?1. Conclusion The predicted value from response surface regression model was matched the measurement data. It indicated that the model showed good fitting degree. This method could be used to optomize the processing suitation of the similar bioactive componds. |
