Abstract: Objective To investigate the extracellular polysaccharides from the saline-alkali plant endophytic fungus SXH-69. Methods The extracellular polysaccharides were isolated from the culture medium of the saline-alkali plant endophytic fungus using ethanol precipitation, Q-Sepharose Fast Flow anion-exchange and Sephacryl S-100 gel-permeation chromatography. The chemical compositions and structural characteristics of the extracellular polysaccharides were studied by high gel permeation chromatography (HPGPC), pre-column high performance liquid (HPLC) derivation, Fourier-transform infrared (IR) spectroscopy and gas chromatography-mass spectrometry (GC-MS). Results Two polysaccharides, sx1-1 and sx2-1, were obtained from the culture medium of the saline-alkali plant endophytic fungus, and their molecular weights were about 13.5 and 18.3 kDa, respectively. The two polysaccharides were mainly composed of mannose. Minor amounts of glucose and galactose were also found. Methylation analysis showed that the extracellular polysaccharides sx1-1 and sx2-1 were mainly composed of terminal Man, 1,4)-Man, 1,6)-Man and 1,2,4)-Man, with some amounts of terminal Glc, 1,2)-Gal, 1,6)-Glc and 1,3,6)-Glc for sx1-1, and sx2-1 with 1,2)-Glc and 1,2,6)-Gal. Conclusion The two extracellular polysaccharides from the plant endophytic fungus SXH-69 was firstly isolated and investigated. The two extracellular polysaccharides had different chemical characteristics, and were heteropolysaccharides with novel structures. |
