      【目的】对中国南海海绵Cinachyrella australiensis来源的一株胰岛曲霉Aspergillus insulicola MD10-2 产aspochracin的发酵培养基进行优化以提高其产量。【方法】依据Plackett-Burman设计对选定的基础培养基进行重要因素筛选,通过单因素实验确定其中心点浓度,再应用响应曲面法拟合后得出最终优化的培养基配方。 【结果】 胰岛曲霉A. insulicola MD10-2 的优化发酵培养基配方为:葡萄糖15.54 g,甘露醇20 g,麦芽糖20 g,谷氨酸钠10 g,玉米浆1 g ,KH2PO4 0.5 g,40.95%浓度人工海水1 L。优化后aspochracin产量达到2.25 mg/g DCW,是原培养基发酵的2.15倍。【结论】通过对胰岛曲霉A. insulicola MD10-2 发酵培养基进行优化可显著提高aspochracin产量。
English Summary:
      [Objective] In the present study, a variety of methods were used to optimize the fermentation medium for aspochracin production by an Aspergillus insulicola MD10-2 derived from the South China Sea sponge Cinachyrella australiensis. [Methods] After the selection of a suitable basic medium , important factors of the basic medium were selected according to Plackett-Burman design. Then the center points’ concentration of the chosen important factors were determined by the single factor experiment. Finally Response Surface Methodology was applied to get a optimized medium formulation for aspochracin production. [Results] Optimized medium formulation is : glucose 15.54 g, mannitol 20 g, maltose 20 g, sodium glutamate 10 g, corn steep liquor 1 g, KH2PO4 0.5 g, 40.95% concentration of artificial seawater 1 L. After the optimization, the yield of aspochracin was increased to 2.25 mg / g DCW, which was 2.15 times of the basic medium. [Conclusion] The yield of aspochracin could be increased by the optimization of fermentation medium remarkably.
