目的 对分离自红树植物海马齿(Sesuvium portulacastrum)的一株内生真菌BY5进行鉴定,并对其发酵产物的生物活性进行研究。方法 采用形态学、28S rDNA序列对红树内生真菌BY5进行鉴定;采用滤纸片琼脂扩散法、自由基清除法 (DPPH、?OH和ABTS ) 和MTT法对其发酵液上清乙酸乙酯抽提物的抗菌、抗氧化和抗肿瘤活性进行研究。结果 菌株BY5的菌落形态和显微形态与曲霉属的基本相同,其28S rDNA序列与塔宾曲霉(Aspergillus tubingensis)的相似度达100%,确定该菌株为塔宾曲霉;其发酵液上清乙酸乙酯抽提物对金黄色葡萄球菌、枯草芽孢杆菌有较强抑制作用;对DPPH自由基、?OH自由基和ABTS 自由基具有较强清除作用(0 mg/mL﹤EC50﹤10 mg/mL);对宫颈癌Hela细胞和人肝癌HepG2细胞具有较强的抑制作用,其IC50值分别为4.44 μg/mL和35.91 μg/mL。结论 红树内生真菌BY5能够产生抗菌、抗氧化和抗肿瘤活性物质,是开发抗菌、抗氧化和抗肿瘤药物的重要资源。 |

Abstract: Objective Identification and biological activities of the strain BY5 from mangrove plant Sesuvium portulacastrum were studied. Method Morphological identification and 28S rDNA sequence were assayed on mangrove endophytic fungus BY5. Using the filter paper disc agar diffusion method, the radical scavenging methods (DPPH ,?OH and ABTS ) and MTT assay, the antibacterial, antioxidant and antitumor activities of the ethyl acetate extract of the fermentation broth were studied. Results The colony morphology and microscopic morphology of the strain BY5 corresponded to the characteristics of Aspergillus sp., and the 28S rDNA sequences showed 100% homogeneous to Aspergillus tubingensis, so the strain BY5 was Aspergillus tubingensis. The ethyl acetate extract of the fermentation broth showed strong inhibition to Staphylococcus aureus, Bacillus subtilis, strong scavenging effect (0 mg/mL ﹤ EC50 ﹤ 10 mg/mL) to DPPH radical, ?OH radical and ABTS radical, and significant inhibition against HeLa and HepG2 tumor cells with IC50 4.44 μg/mL and 35.91 μg/mL, respectively. Conclusion Mangrove endophytic fungus BY5 was important resource producing rich antibacterial, antioxidant and antitumor substances. |
