目的 分离筛选黑海参中具有抗肿瘤活性的三萜皂苷。方法 采用萃取法、大孔树脂和正相硅胶色谱分离对黑海参三萜皂苷进行提取分离,以MTT法检测肿瘤细胞活性,PI染色及Hoechst 33258染色检测肿瘤细胞周期及凋亡。结果 分离获得A-I 9个组分,其中C、E、F组分对人肺癌细胞A549和白血病细胞株HL60显示明显的抗肿瘤活性,而对肝癌BEL7402和MOLT-4细胞无影响。结论 利用抗肿瘤筛选模型进行跟踪分离黑海参中有效部位,对进一步深入分析开发有价值的抗肿瘤先导化合物提供了科学依据。 |
English Summary: |
Objective Separate and identify triterpene saponins from the sea cucumber Holothuria atra Jaeger with antitumor activity. Method By extraction, macroporous resin and silica gel normal phase chromatography, the saponins from sea cucumber Holothuria atra Jaeger were preliminarily isolated. The cell viability was detected by MTT, cell cycle and apoptosis were measured by PI and Hoechst 33258 stain respectively. Results Saponins from sea cucumber Holothuria atra Jaeger were separated into 9 parts, named as A-I. C, E and F decreased cell viabiltys of A549 (human lung cancer cells) and HL60 (Leukemia cell lines) significantly but showed no effect on BEL7402 and MOLT-4(liver cancer cells). C induced G0/G1 cell cycle arrest and led to apoptosis in A549. Conclusion The study on separation of the effective parts from the sea cucumber Holothuria atra Jaeger with the antitumor screening model will provide scientific basis to develop valuable antitumor lead compounds. |
