      应用全细胞膜片钳制技术观察了羧甲基甲壳胺对豚鼠单一心室肌细胞钙离子电流(I_(ca))的作用。实验结果显示:羧甲基甲壳胺能抑制心室肌细胞的Ca~(2+)内流,使I_(ca)减小。此作用具有明显的电压依赖性,对其反转电位无明显影响。羧甲基甲壳胺对I_(ca)的作用也具有明显的浓度依赖关系,浓度为0.05%、0.1%、0.2%的羧甲基甲壳胺使I_(ca)分别减小11. 8%、30%和 48.1%,最大效应浓度接近 0. 25%。此外,停止给予羧甲基甲壳胺后,其对I_(ca)的作用减小直至消失,I_(ca)可基本恢复至给药前的水平。上述结果提示,羧甲基甲壳胺减小Ca~(2+)内流的作用可能与其杭心律失常作用有关。
English Summary:
      With patch clamp method,the effects of CM-Chitosan on slow inward calci um current (I_(ca))were investigated in single ventricular cells of guinea pigs. The results shown that CM-Chitosan decreased I_(ca) in a concentration and voltage-dependent mode,but had lit tle effect on reversal potential. CM-Chitosan 0. 05 % decreased I_(ca) by 11. 8 % and its maximal effectie concentration was near 0. 25%. Above all suggested that CM-Chitosan Could block I_(ca) which might be one of the antiarrhythmic mechanisms of CM-Chitosan.
