目的 对采自西沙群岛的软珊瑚样品Sinularia sp.进行倍半萜类次级代谢产物的研究。方法 利用硅胶柱层析、薄层色谱、中压制备型液相色谱和高效液相色谱等分离方法,对软珊瑚中的化学成分进行提取、分离并纯化;通过计算ECD、X射线单晶衍射方法确定新化合物1~2的绝对构型,综合运用核磁共振波谱、质谱等现代波谱学方法,通过与文献报道的数据进行对比,鉴定已知化合物3~8的结构;结果 从西沙群岛软珊瑚Sinularia sp.中分离鉴定8个倍半萜类化合物,包括2个新的倍半萜sinularnoid A (1)、sinularnoid B (2),和6个已知的倍半萜类化合物,分别为sinuhirtin F (3)、nanonorcaryophyllene B (4)、sinuhirtin G (5)、nanonorcaryophyllene A (6)、methyl-(1S*,4S*,5S*,9R*,11S*)-4,5-epoxycaryophyllen-13-oate (7)、(1R,4R,5R,9S,11R)-4,5-epoxycaryophyll-8(14)-en-12-oic acid (8)。结论 从西沙群岛软珊瑚Sinularia sp.中分离得到8个倍半萜类化合物,其化学结构类型分别属于caryophyllene型和norcaryophyllene型,其中化合物1~2为新化合物。活性测试结果表明,化合物7在斑马鱼实验中具有微弱的抗炎活性。 |

Objective To investigate the secondary metabolites of sesquiterpenoids isolated from a soft coral Sinularia sp. collected from the Xisha Islands. Methods The chemical components of soft coral were extracted, isolated and purified by silica gel column chromatography, TLC, MPLC and HPLC. The absolute configuration of new compounds 1~2 was confirmed by calculated ECD, X-ray diffraction analysis, and the structures of known compounds 3~8 were determined by extensive spectroscopic analysis, including NMR, MS, and by comparison of the spectral data with those previously reported in the literature. Results Eight sesquiterpenoids were isolated from Sinularia sp., including two new sesquiterpenoids, namely sinularnoid A (1), sinularnoid B (2), together with six related known analogs, sinuhirtin F (3), nanonorcaryophyllene B (4), sinuhirtin G (5), nanonorcaryophyllene A (6), methyl-(1S*,4S*,5S*,9R*,11S*)-4,5-epoxycaryophyllen-13-oate (7), (1R,4R,5R,9S,11R)-4,5-epoxycaryophyll-8(14)-en-12-oic acid (8). Conclusion Eight sesquiterpenoids with caryophyllene and norcaryophyllene-type skeletons were isolated from the soft coral Sinularia sp. Compounds 1~2 were new compounds. Compound 7 showed weak anti-inflammatory activity in zebrafish assay. |
