      目的 研究一株来源于海南东寨港红树林植物无瓣海桑的内生真菌Aspergillus niger SAF17次级代谢产物及其抗菌活性。方法 运用硅胶柱层析、凝胶柱层析及高效液相色谱等分离手段对SAF17乙酸乙酯粗提物进行单体化合物的分离纯化,并采用微量稀释法测定化合物的体外抑菌活性。结果 基于波谱学数据分析和文献数据对比,从SAF17乙酸乙酯流浸膏中分离鉴定出12个化合物:campyrone A (1)、campyrone B (2)、campyrone C (3)、(22E,24R)-9α,15α-dihydroxyergosta-4,6,8(14),22-tetraene-3-one (4)、3β-Hydroxyl-(22E,24R)-ergosta-5,8,22-trien-7,15-dione (5)、(+)-pestalazine A (6) 、(+)-iso-pestalazine A (7)、 asperazine (8)、fonsecinone A (9)、aurasperone A(10)、asperpyrone B(11)和asperpyrone C(12);其中化合物1-3为吡喃酮类化合物,化合物4-5为麦角甾醇类化合物,化合物6-8为肽类化合物,化合物9-12为二聚萘并吡喃酮类化合物。抗菌活性实验表明,化合物3对耐甲氧西林金黄色葡萄球菌的IC50值为96.47μg/mL,化合物2、3、9和10对大肠杆菌的IC50分别为82.14、94.75、95.15和77.19 μg/mL。结论 菌株Aspergillus niger SAF17可以产生的化合物种类丰富;化合物4、6和7为首次从该菌种分离得到;化合物3对耐甲氧西林金黄色葡萄球菌具有抑菌活性,化合物2、3、9和10对大肠杆菌具有抑制作用。
English Summary:
      Abstract:Objective The secondary metabolites of Aspergillus niger SAF17, an endophytic fungus derived from the mangrove plant sonneratia apetala in Dongzhaigang, and their antibacterial activities were investigated. Methods The ethyl acetate crude extract of Aspergillus niger SAF17 was subjected to various separation techniques, including silica gel column chromatography, gel column chromatography and high performance liquid chromatography (HPLC). The antibacterial activities of those pure compounds were determined by microdilution method. Results Twelve compounds were obtained, and their structures were identified as campyrone A (1), campyrone B (2), campyrone C (3), (22E,24R)-9α,15α-dihydroxyergosta-4,6,8(14),22-tetraene-3-one (4), 3β-Hydroxyl-(22E,24R)-ergosta-5,8,22-trien-7,15-dione (5), (+)-pestalazine A (6), (+)-iso-pestalazine A (7), asperazine (8), fonsecinone A (9), aurasperone A (10), asperpyrone B (11) and asperpyrone C (12) based on NMR and MS data, and comparing with literature data. Compounds 1-3 are pyranones, compounds 4-5 are ergosterol, compounds 6-8 are cyclic peptides, and compounds 9-12 are dimeric naphthopyranones. The antibacterial experiments showed that the IC50 value of compound 3 against methicillin-resistant Staphylococcus aureus was 96.47 μg/mL, and compounds 2、3、9 and 10 against Escherichia coli were 82.14, 94.75, 95.15 and 77.19 μg/mL, respectively. Conclusion Aspergillus niger SAF17 can produce a variety of compounds. Among them, compounds 4, 6 and 7 were isolated from this strain for the first time. Compound 3 displayed weak inhibitory activity against methicillin-resistant Staphylococcus aureus, and compounds 2、3、9 and 10 possessed weak antibacterial activity against Escherichia coli.
