| 目的 研究氟苯尼考纳米乳在大鼠体内的药代动力学行为。方法 利用高效液相色谱-质谱联用(LC-MS/MS)技术建立检测氟苯尼的分析方法,并进行方法学验证。分别灌胃给予大鼠氟苯尼考纳米乳和市售可溶性粉,收集不同时间点的血浆样品,测定血浆样品中的氟苯尼考浓度,计算AUC等药代动力学参数,考察氟苯尼考纳米乳在大鼠体内的药代动力学特征。结果 以20 mg/kg的剂量单次口服给药氟苯尼考纳米乳和市售氟苯尼考可溶性粉后,AUC0-10h分别为27.5 ± 6.1和25.9 ± 1.1 h*μg/mL,AUC0-∞分别为29.1 ± 6.6和26.2 ± 1.1 h*μg/mL。结论 制备的氟苯尼考纳米乳较市售氟苯尼考可溶性粉在大鼠体内的AUC0-10h和AUC0-∞分别提高6.2%和11.1%,一定程度上可以改善氟苯尼考在动物体内的药动学行为。 |

| Objective To investigate the pharmacokinetic characteristics of flufenicol nano emulsion in rats. Methods A high-performance liquid chromatography-mass spectrometry (LC-MS/MS) detection method for flufenicol in plasma was established and verified. Plasma samples were collected at different time points after intragastric administration of flufenicol nano emulsion and flufenicol soluble powder. The concentration of flufenicol in plasma samples was measured to calculate the AUC and other pharmacokinetic parameters, and the pharmacokinetic characteristics of flufenicol nano emulsion in rats were investigated. Results After single oral administration of flufenicol nanoemulsion and commercially available flufenicol soluble powder at 20mg/kg, AUC0-10h were 27.5±6.1 and 25.9±1.1 h*μg/mL, AUC0-∞ were 29.1±6.6 and 26.2±1.1 h*μg/mL, respectively. Conclusion The AUC0-10h and AUC0-∞ of flufenicol nanoemulsion in rats were increased by 6.2% and 11.1%, respectively, compared with that of commercially available flufenicol soluble powder, which could improve the pharmacokinetic behavior of flufenicol in animals. |
